Are you on Windows XP?

Try Control Panel>Sounds and Audio Devices>Audio, and check that your MIDI Playback Device is set to Microsoft GS Wavetable SW Synth, and then click Volume (just below it) and make sure that the SW Synth Fader is turned up, as well as your WAVE and Master Volume faders.
Look at ur mixer device properties too the in and output.....see how its set up .........make sure there's nothin off that should be on or muted

try that and if that dont work try uninstalling gp5 and when u are re-installing it check wut is gunna play for your midi play back..........
 
rebooting your computer helps after you change your computer settings
 
Hate to bump this again but...

It seems i've tried everything, including reinstalling the program, rebooting, etc. nothing is muted. Even after a reinstall, the same problem is still occurring.
 
I just downloaded and installed something called tux guitar, which seems to be basically the same program for free, and it does the EXACT SAME THING as guitar pro 5.
