It shoundn't make and difference, or maybe I should say not enough difference to hear. Do take the grill off before painting it though and use as light of a coat of paint as possible. If you are going to a darker color you might consider using permanent ink applied with an airbrush or other small sprayer.
Thanks y'all. Yes I would definitely have the grill cloth off before spray painting.
And I'm talking about spray painting the whole thing, not just a patch. It's just an idea, but it's for an amp that I'm building, and I think the grill cloth would look good as a certain color that I can't seem to find anywhere.
Be careful, certain types of grille clothe will stretch when you get them wet. You can sometimes shrink them with a hairdryer, but not always. A freind of mine wanted to remove some stains from an old Vox amp and was sorry he tried.

I am almost certan that the grille cloth will be painted with it off of the cabinet.
once the paint dries and the cloth is installed it can be stretched tight so it does not sag.
also the grille cloth on that vox can be removed then re installed to remove any sags in the material.
