Basically converts line signal (instrument) to mic signal. Usually 1/4" to XLR. Stands for Direct Injection.
 
Their purpose is one or all of three:

1. Impedance matching
2. Balanced(3-conductor) to Unbalanced(2-conductor) conversion
3. Earth Ground Lift/Connect

For most uses it converts an unbalanced, high impedance signal (the kind of signal generated by most pickups and dynamic mics) into a balanced, low impedance signal (the kind of signal required by most mixers and pre-amp inputs). It also isolates the output signal from the input signal, so it can be used to cure ground loop problems in other parts of the signal chain (like between a mixer output and a power amplifier input).

just to be exact, Line level and instrument level are not the same thing.
 
DI (Direct Injection) boxes are designed to take an unbalanced low- to medium-level, high-impedance signal
from a bass,keyboard etc and convert it to a mic-level, balanced and low-impedance signal intended for
connection to a sound desk microphone input/mic pre. Most DI boxes also have an earth-lift facility to help break ground loops.

There are two basic types of DIs: Active and Passive
Passive DI's use a transformer to convert the electrical signals while
active direct boxes use an amplifier circuit. Passive DI's have the advantage of providing isolation
which can reduce ground hum in systems.
 
SOMME DI boxes also allow you to take the speaker output of a guitar amp and bring it down to mixer level.
