Hi, just received an acoustic guitar in the post which was bubble wrapped, cardboard wrapped and in a box. I've undone the guitar already seeing as the packaging wasn't offering much insulation to around room temp 20'C - should I wait before tuning it up and if so how long? Thanks

By the time you read this, or any other post here, the guitar will be at room temp. Go right ahead and tune it up!

Out of curiosity, how much does temperature/humidity/weather I general affect the tuning of a guitar? Will an in tune guitar go out of tune on an hour long car ride in really hot/cold/humid weather?

Some guitars/strings do, some don't! A friend could not keep his Strat with new strings in tune at a club where the a/c was blowing right at his side of the stage, even after a couple hours of acclimating.

Every guitar/situation is different. The best way to test this is to take it places, play it, don't worry about the weather or temperature or humidity and tune it whenever it's out of tune :)

Unless of course it's raining or a million degrees or 99% humidity etc.
