Do these really work?

I haven't used the Alesis unit but I have used a few software programs that remove vocals. A lot of it depends on the material and how the song is recorded. First, the vocals must be a stereo track. The unit probably does a few things depending on the preset. The most basic is inverting the phase of one side. If the vocals are balanced, this will remove them by cancellation. It probably also uses eq to zap certain frequencies. Also if there is a lot of reverb or delay, that may not be removed.

Like I said, a lot depends on the recorded material you are working with. Some songs it will work great and others, not so much.

My Digitech JamMan looper has a remove-vocals/lead guitar feature, and it only cuts them down- the vocals or lead guitar is still quite discernable. It HELPS for learning- I can't lean on the vocals/led guitar as much, but I would never use those loops as backing tracks.
