So i just got some new equipment, a Tascam US-144 MK:II, which has Cubase LE 4 software included. Everything installed fine, but I have no idea where to start. I tried recording my guitar on audio and instrument tracks (plugged into the GUITAR IN port on the Tascam) , and i'm not getting any sound off of my computer. HELP!

A couple of thoughts as to why there is no sound.
On most DAW's (I have no experience with Cubase) you have to tell your program in the preferences section where to look for your audio input. If your tascam is plugged in to the USB port, it should be listed as a choice. Select that if you haven't done that already.
I assume you are playing a guitar with a pick-up going directly in to the line in, with the tascam set on guitar. If you are micing the guitar and going in to the XLR input, you need phantom power on to your mic if it's a condenser mic, and the tascam should be set to mic. Which way are you recording the guitar? Then all you need to do is create a guitar track in Cubase, arm it for recording and hit record. If you've done that, are you getting a waveform in Cubase with no sound or nothing at all? If you are getting a waveform, the sound is getting in to Cubase and the problem is with your monitoring. It may be as simple as hitting the "monitor on" switch. Give us more info and we can probably help figure it out.

Yes, i did notice if i open a new instrument track I can select the Tascam as a source, yet i'm still not getting a reading.
The guitar is plugged straight into the Tascam, via Guitar input, it is not miked. I also got the monitor suggestion from the quickstart manual, but that didn't seem to work either...

The tascam has two channels a right and a left. Make sure you have selected the same channel to record as the guitar is plugged in to, i.e. if you have your guitar plugged in to the right channel, and the track is armed to record the left channel, you'll get nothing. Have you tried the other channel in Cubase?

Okay, update. I got Cubase to recognize my guitar, and it is recording, so thank you very much for the help. Only problem is now...when i play it back, there is no audio....?

You need to set the relevant output bus to the Tascam interface on the relevant tracks. You can check your i/o setup by pressing F4.

a little more specific?

You have to select the Tascam as your output, just like you did the input. Them make sure you've got your monitors/phones plugged into the Tascam correctly. What are you using to monitor with? Powered monitors? Phones?

a little more specific?

If you press F4 you can see and configure buses. You need to check where these buses route to and whether your track is routed to the main out (which chances are it will be but the VST connections window should help to check your software routing setup).
