I would treat an axe-fx input that same as a gtr amp input. FWIW, I have used a Little Labs redeye for years and it does double duty as a DI and reamp box.

Remove the tube cover and gently wiggle them in the socket. Make sure they are seated all the way in the socket. Then wipe the tube off with a cloth. Exercise the Loop Jack is take a plug and insert it remove it several times fast.

That's probably a cold solder joint. As the amp heats up and thermal expansions take place, the connect becomes a high resistance point. As a first step re-seat the tubes and exercise any loop jacks. Does tapping on the amp while it's sizzling change it? Cold solder joints are usually...

Gain staging is ensuring the interface levels/types are correct or match A gtr DI box is a gain stage moving the gtr level to a mic level. The same when reamping going from whatever level you're output is to an amp input level.
