I'm pretty sure you have to bounce the track down with the effect, essentially "printing" the track with the effect applied. I have a Neo but don't use the internal effects modules much.

I think auto-tune has a place if it's applied with extreme care or used as a studio effect/gimmick. It really bugs me when it pops up in places in shouldn't. Check out Dust Bowl Children (track 2) on the latest Alison Krauss...

Another +1 for the 2488. I just got mine and it's wonderful, especially after experiencing a ton of computer woes with my old setup. Down the line, I want to go back to a computer-based DAW, but will likely use the 2488 on the front end to record (I just like having real knobs and sliders)...
