    I would buy 'mastering audio' by Bob Katz, in that book he has a chart called the 'musical pitch relation chart' it helps to understand what frequencies to boost and cut, still nothing beats experience.

    there's no reverb, way too dry, the other thing is I can't hear a solid bass drum, the arrangement could be improved by adding strings at 1:04, and at 0:50 in the breakdown section, I'd have that as a brass arrangement instead of organ, to help keep it interesting.

    yes you are spot on, sustain is super important, it's where 'the pocket' of the groove is. Sometimes I shorten notes, and sometimes lengthen them slightly, using gates can be interesting as well in how it affects groove.

    I edit them quite a bit, sometimes to make notes shorter to help the groove, obviously it's always better when a bassist can actually play perfectly though.
