On the far left bottom corner is a little metal tab. You can clip your probes to that for testing. It says you should see 2v p-p @ 1khz. (p-p means peak to peak. Very top of sine wave to very bottom)
What is giving you the error message? Reaper? or Windows? The sound card shouldn't make a difference if you are trying to send MIDI data to the keyboard, Be sure to read the manual to fully understand how to use MIDI.

For sure it's unbalanced, though the manual doesn't specify. But effects loops are designed so you can plug stomp boxes into them, which are unbalanced. The manual does caution to use short, well-shielded cables. That warning wouldn't be necessary if the connections were balanced.
