I've got a 4 inch speaker for my guitar amp. I'm thinking of taking it off and adding on a 8 inch speaker. Would this work? Would this do anything? Why or why not? Thanks!

yeah you could do this, you would just half to cut a bigger whole. It would make a difference (most likely) in that a speaker twice as big as your old one would probably have more wattage. I'm not an expert on how a speaker with more wattage changes the volume of the same amp but i know first hand that it does.
 
The speaker might handle more watts, but the amp would be unchanged.

Ideally you want to match the impedance before and after. If 8 ohms before, you want an 8 ohm for the larger speaker.

Larger speakers push more air and generally handle bass better. It may or may not be louder depending on other factors like speaker efficiency.

I did this with a small Fender amp going from a 10" to a 12". Worked well for me.

Does it change the volume at which the sound starts to distort on a tube amp?
 
Depends.

If distortion was coming from the speaker, then it well might sound better.

If coming from amp, it boils down to how efficient the new larger speaker is compared to the older smaller model. If more efficient, then it would produce more volume for the same amp setting, and hense distort later in the amp volume cycle.
